Technical Note - 10x Barcode Continuity during Sequencing of Genome Libraries

This Technical Note describes the importance of capturing the first 16 cycles of Read 1 during the sequencing of 10x Chromium Genome libraries to ensure the complete 10x Barcode is sequenced. If the sequencing run is aborted during the initial cycle and then resumed (e.g., HiSeq® 4000 and HiSeq® X Series sequencers have this capability), the information for the first base will be lost when the run is aborted and only the last 15 bases of the 10x Barcode will be captured. If the complete 10x Barcode sequence is unavailable, the fragment cannot be assigned to the correct partition and the library will need to be resequenced.
